So I'm always doing something with a rune as Runos. I never thought of it until now but I thought I could probably write up the theory behind it so it makes more sense, especially since Siivyra has asked if she could be instructed in the arts.
Now, most of the time I just try to make things up as I go, but I try not to be evilly overpowered about it. If I fight someone, chances are you'll get tagged with an inscription but I will always let you decide if it works or not.

Most runes only work in reaction to the runic hide curse, a complex spell bound to Runos' soul that burns onto the flesh of the vessel containing it to keep it bound. When the body is utterly destroyed, the soul will linger in the spot a moment, then forcibly latch onto the nearest body, sometimes even able to reach between worlds and planes to grab one if others are not close enough. The previous owner's soul will either be subdued until the soul expires, or the soul will be expelled (unless the individual has a strong will, in which case they may share the body like Azaraen). The spell originally was developed by a wizard gone insane, using Runos as his test subject (He botched it on himself when he was content and exploded). The original rune pattern was pretty unstable, and Runos in the course of her life has exploded more than once, leaving devastation, and taking another body (it is entirely possible Runos has been more than just a dragon over many lives, but she wouldn't remember what she started out as). Overall each rune has been arranged for a purpose, some regulating body functions like breathing, others are binders to make sure two runes will work together.

Think of it as like speaking a language. As in language, which is composed of many words, you will have some basic words. Here you have the base runes, which are for things like elements. Of them, the list is small, composing of runes such as Fire, Water, Earth, Air, Cold, Energy, Life, Death, Spirit, and there is a unique rune called the anchor or the binding rune that holds everything together.
Now when you begin to look at these runes and begin to derive 'words' out of them, you get new 'words' or rather derived runes. For example, a spell I like to not use on people often unless they have caused some annoying trouble is "Burning Obsidian"; they become heavily encased within exactly that. It is composed of forms similar to fire and earth, and another variant has the obsidian always restoring itself if they struggle to break pieces of it off by adding life...
One rune not often used is the death rune, as it is very dangerous, unstable, and does not work with many other runes.
Another similarity to language is grammar, there is always an order and a way to do it, and consider how the rune is shaped to be how you pronounce it. There is only one way to pronounce a word sometimes. :)
And lastly, the words themselves. Sometimes words do not exist until the word is invented, while other words simply will never exist as hard as you try. The runes can do many things but.. always they are limited to the vocabulary of the language.

About forms I've used:
Obsidian Prison: Uses earth variant with the anchor variant to make it stick around the target. Add life for a creeping prison. Add fire variant for burning obsidian prison.
Anchored Teleport: Uses Air and the Anchor. You can teleport things to something you have a focus over, or you can anchor it to a marker rune. Add spirit to do runos' old "Leave the statue" trick (which doesn't work anymore because she does not occupy a statue anymore).
